Effects of retinoids on proliferation of human melanoma cell line A375
【Abstract】 AIM: To investigate the effects of diverse retinoids and receptor agonists in inhibiting proliferation of human melanoma cell line A375. METHODS: Human melanoma cell line A375 was treated by diverse retinoids (atRA, 13cis RA and 9cis RA), MA (RXR agonist) and TTNPB (RAR agonist) and their effects on the proliferation and cell cycle of A375 cells were analyzed by MTT and flow cytometer. RESULTS: Both retinoids and TTNPB inhibited the proliferation of A375 cells, but MA had no such effect. At all concentrations (10-5, 10-6 and10-7 mol/L) in 24 h and 10-5 mol/L in 48 and 72 h, TTNPB had the most powerful effect in all reagents (P<0.05). The results of cell cycle analysis showed that retinoids and TTNPB caused the blockage of G0/G1 phase, but MA had no such effect. At 10-5 mol/L in 24 and 48 h, TTNPB had the most powerful effect of causing blockage of G0/G1 phase in all reagents (P<0.05). CONCLUSION: Retinoids can inhibit the proliferation and cause the blockage of G0/G1 phase in human melanoma cell line A375. It is not the activation of RXR but the activation of RAR that may be related to these effects.
